Lactate dehydrogenase (LDH) is an important indicator in clinical myocardial enzyme spectrum analysis and can be used for the auxiliary diagnosis of heart disease. Since LDH is released during tissue damage (usually due to loss of cell membrane integrity), it is also considered a biomarker for cell necrosis, injuries, and related diseases, and is widely used in cytotoxicity assay.
There are various colorimetric methods for measuring LDH activity, such as the DNP method, INT method, WST-8 method, etc. The DNP method is relatively cumbersome and less commonly used. The INT method, such as the Beyotime LDH Cytotoxicity Assay Kit (C0016/C0017), is based on the reduction of NAD+ to NADH under the action of LDH, and the subsequent reaction of NADH with INT (2-p-iodophenyl-3-nitrophenyl tetrazolium chloride) catalyzed by diaphorase to generate NAD+ and formazan.
The latest launched LDH Cytotoxicity Assay Kit with WST-8 (C0018) uses the WST-8 method, which has higher sensitivity, greater absorbance changes, and more accurate results compared to the INT method.
Working Principle
Under the action of LDH, NAD+ is reduced to NADH, and NADH reacts with WST-8 to generate NAD+ and water-soluble formazan dye, which produces an absorbance peak at 450nm wavelength (Figure 1). This allows for the quantification of LDH activity through colorimetric analysis, where absorbance is linearly correlated with LDH activity.

Product Features
● Easy operation: The reagents in this kit do not harm the cells, so there is no need to remove the cell culture medium. The working solution can be directly added to the cell culture well. Alternatively, cell culture medium can be taken for assay, allowing the cells to be used for other experiments.
● Good stability: Although the working solution is recommended to be prepared freshly, it can also be stored at 4℃ for up to 3 days.
● Wide application: This kit can detect D-LDH and L-LDH, or a mixture of both. The sample can be cell culture, cell lysate, etc.
