Serum-free B27 Supplements

The growth maintenance effect of Beyotime B27 Supplement (50X, Serum-free) (C0350) on primary mouse neuronal cells and the immunofluorescence staining effects using DAPI staining solution and Tuj1 antibody:

Beyotime newly launches four serum-free B27 supplements, widely used to support the growth and differentiation of hippocampal and other CNS neurons, maintain short- or long-term viability, induce embryonic stem cells to differentiate into neural lineage cells, and culture pluripotent stem cells.

Selection Guide:

B27 Supplement(50X,Serum-free)(C0350) is a complete formula for long-term growth and maintenance of neuronal cells.

B27 Supplement(50X, Minus Insulin)(C0348) does not contain insulin and is recommended for use in studying the growth state of hippocampal and other CNS neurons with supplementation of insulin.

B27 Supplement (50X, Minus Anti-oxidant) (C0349) does not contain any antioxidants and is the primary choice for studying oxidative damage, cell apoptosis, and neuronal free radical damage.

B27 Supplement(50X, Minus Anti-oxidant)(C0347) has no vitamin A that induces neuronal cell differentiation, and is an ideal choice for the growth and proliferation of neural stem cells.

Base Medium Selection:

C0350 works with Neurobasal and Neurobasal-A media for normal culture, growth, and differentiation of neuronal cells.

C0347/C0348/C0349 with Neurobasal Medium support the growth and viability of prenatal and embryonic neurons.

C0348/C0349 with Neurobasal-A Medium are suitable for long-term survival and viability of neonatal and adult primary brain neurons.

C0349 with DMEM/F12 supports serum-free growth of CNS, PNS, embryonic and adult neural stem cells (NSCs) or neural progenitor cells.

B27 serum-free supplement VS N-2 serum-free supplement:

B27 supplement is mainly used for the growth and maintenance of neural stem cells and neuronal cells, while N-2 supplement is mainly used for culturing neuroblastoma or post-mitotic neuronal cells from PNS/CNS.

For low cell seeding density, it is recommended to use a combination of B27 and N2 for culturing neural stem cells and neurons.

For high cell seeding density, N2 is recommended for culturing neurons and glioma stem cells.

N2 can be combined with B27 for serum-free culture after directed differentiation of pluripotent stem cells.

Precautions

  1. We recommend thawing this product overnight at 2-8℃ or at room temperature for 2 hours to ensure its optimal performance.
  2. Once B27 is added to the culture medium, please use it within 2-4 weeks to maintain its effectiveness in supporting neuronal cell growth.
  3. As shown in the figure below, we recommend supplementing L-glutamic acid to a final concentration of 25μM in Neurobasal medium containing 1X B27 Supplement and L-glutamine for the initial cultivation of primary neuronal cells. After 4 days of culture, the medium needs to be replaced as primary neuronal cells need to be cultured for a long period of time under L-glutamic acid-free conditions.
