Working Principle
CM-H2DCFDA itself is non-fluorescent and can freely cross cell membranes. However, upon the cell entry, it is hydrolyzed by intracellular esterase to produce CM-H2DCF (also known as CM-DCFH), which cannot permeate the cell membrane. The chloromethyl group of CM-H2DCF can react with intracellular glutathione or other thiols, enhancing its retention in cells. Intracellular ROS can then oxidize the non-fluorescent CM-H2DCF to produce fluorescent CM-DCF. The fluoresence intensity is proportional to the intracellular ROS levels.
