| Background | Activin A, a multifunctional cytokine belonging to the TGF-β superfamily, forms dimers via disulfide bonds linking two beta-A chains. It regulates diverse physiological processes including stem cell maintenance and differentiation, hypothalamic-pituitary-gonadal (HPG) axis function, germ cell maturation, erythropoiesis, insulin secretion, neuronal survival, embryogenesis, and osteogenesis. Activin A binds to cell surface receptors on embryonic stem cells and induces the expression of endoderm-specific genes including Gsc, Sox17 and Mixl1, which are capable of inducing the differentiation of embryonic stem cells into definitive endoderm cells [1, 2]. |

图1. 碧云天BeyoES™ Human Activin A (C0321/C0322)在还原(R, Reducing)及非还原(NR, Non-Reducing)条件下的SDS-PAGE电泳检测效果图。本产品经BeyoGel™ Elite PAGE预制胶(Tris-Gly, 4-20%, 15孔) (P0822)电泳，分子量标准为BeyoColor™彩色预染蛋白分子量标准(10-170kD) (P0075/P0076/P0077)，并经BeyoBlue™考马斯亮蓝超快染色液(P0017F)染色。结果显示，本产品在还原条件下为单体(Monomer)，分子量约为15kDa；在非还原条件下为二聚体(Dimer)，表观分子量约为26kDa。实际检测结果可能会因样品和检测条件等的不同而存在差异，图中数据仅供参考。
本产品为粉末形式，经过除菌处理，加入适量细胞培养级超纯水(ST875)充分溶解后可以直接用于细胞培养实验。
本产品可以用于人胚胎干细胞或多潜能干细胞的诱导分化。本产品用于诱导H9人胚胎干细胞分化48小时后的细胞形态和生长状态请参考图2。

本产品诱导H9人胚胎干细胞分化5天后，通过qPCR检测内胚层分化标志基因AFP、GATA6及SOX17表达水平的效果参考图3。

注意事项：蛋白的每次冻融均会引起部分失活，因此首次配制成相应浓度的储存液或工作液后(请根据产品简介中Reconstitution一栏的信息配制储存液)，须适当分装后-20ºC或更低温度冻存，尽量避免反复冻融。
本产品经过无菌处理，溶解后可以直接用于细胞培养实验，使用过程中请注意无菌操作。
为避免微量蛋白被离心管管盖或管壁非特异性吸附，首次使用建议先适当离心，然后再开启。
