Lipid peroxidation (LPO) is a key change in various pathological conditions, leading to alterations of important membrane protein structure and function, cellular dysfunction, as well as extensive cell and tissue damage, closely associated with the development of many diseases.
Ferroptosis is an iron-dependent cell death distinct from apoptosis. Under the influence of ferrous iron, the antioxidant capacity of cells decreases, leading to the oxidation and destruction of phospholipids including long-chain unsaturated fatty acids on cell membranes or organelle membranes, causing membrane disruption and ultimately regulated cell death (RCD), also known as programmed cell death (PCD). Extensive lipid peroxidation is a key downstream feature and an important indicator of ferroptosis.
Beyotime Lipid Peroxidation Assay Kit with BODIPY 581/591 C11 employs a fluorescent probe to rapidly and sensitively detect the changes of lipid peroxidation levels in live cells. The staining results can be examined by fluorescence microscopy, confocal microscopy, fluorometer, flow cytometry, and other fluorescence detection systems.
Working Principle
BODIPY 581/591 C11 is a fluorescent probe for the assay of lipid peroxidation and antioxidant capacity in living cells. In its reduced state, it has a maximum excitation wavelength of 581nm and a maximum emission wavelength of 591nm, resulting in red fluorescence (high red-to-green fluorescence ratio). Upon oxidation by lipid peroxides, the excitation and emission maxima shift to approximately 488/510nm, leading to green fluorescence (reduced red-to-green fluorescence ratio). Therefore, the degree of lipid peroxidation can be determined by observing the red and green fluorescence using standard settings.
