碧云天生产的Poly(U) Polymerase,即Poly(U)聚合酶,也称PolyU Polymerase或PolyU聚合酶,简称PUP,是由碧云天自主研发的PerfectProtein™技术平台表达、纯化获得的重组聚合酶。Poly(U) Polymerase能以不依赖模板的方式将来自UTP的UMP整合到底物RNA的3'末端,形成poly(U)尾;也可以不依赖模板的方式将来自ATP的AMP整合到底物RNA的3'末端,形成poly(A)尾,但该酶形成poly(U)尾的能力比形成poly(A)尾的能力强得多。如果反应中使用生物素等标记的UTP,则可实现对RNA的3'末端的生物素标记等[1]。

图1.碧云天Poly(U) Polymerase (R7072)和国外N公司的同类产品(Competitor)对RNA 3'末端加Poly(U)尾的效果图。在25µl反应体系中加入图中指定量的本产品或N公司的Poly(U) Polymerase,37ºC孵育1小时进行反应,65ºC加热20分钟进行失活,反应完毕后加入2X RNA Loading Buffer (R0215),95ºC孵育3分钟进行变性,随后进行尿素变性聚丙烯酰胺凝胶电泳。如图所示,本产品与N公司的产品相比,具有类似的对RNA 3'末端加Poly(U)尾的效果。反应体系(25μl):50mM NaCl, 10mM Tris-HCl, 10mM MgCl2, 1mM DTT, pH7.9 @ 25ºC, 0.5mM UTP,人工合成的80pmol的16nt长度的RNA以及不同浓度的Poly(U) Polymerase。实际检测效果会因实验条件、检测仪器等的不同而存在差异,图中数据仅供参考。
用途:RNA 3'端加Poly(U)尾或Poly(A)尾,然后用于克隆;用生物素或荧光修饰的UTP标记RNA;放射性标记RNA;对3'末端为2'氧甲基的RNA进行Poly(A)加尾;对进入真核细胞的RNA加尾从而研究其对RNA稳定性和翻译的影响。
来源:由大肠杆菌表达,表达基因为来自Schizosaccharomyces pombe Poly(U) Polymerase基因。
活性单位定义:One unit is defined as the amount of enzyme that incorporates 1nmol of UMP into RNA in a 50μl volume in 10 minutes at 37ºC.
纯度:不含DNase、RNase和磷酸酯酶。
酶储存溶液:10mM Tris-HCl (pH7.5 @ 25ºC), 100mM NaCl, 0.1mM EDTA, 1mM DTT, 50% (v/v) Glycerol.
失活或抑制:65ºC加热20分钟可使Poly(U) Polymerase失活。
10X Poly(U) Reaction Buffer:500mM NaCl, 100mM Tris-HCl, 100mM MgCl2, 10mM DTT, pH7.9 @ 25ºC.

注意事项:10X Poly(U) Reaction Buffer中不包含UTP。
Poly(U) Polymerase的用量主要影响目的产物的长度,可以根据所需目的产物的长度适当调整酶的用量,如果想得到长度较长的产物可以适当加大酶的用量或延长反应时间。
本产品中在使用时宜存放在冰盒内或冰浴上,使用完毕后宜立即放置于-20ºC保存。
超纯水推荐使用BeyoPure™ Ultrapure Water (DNase/RNase-Free, Sterile) (ST876)。
